Pengaruh Simvastatin Terhadap Kapasitas Fagositosis Makrofag Pada Mencit Balb/C Yang Diberi Lipopolisakharida
ABSTRACT

Background: Simvastatin is included in a group of medicine called hydroxy metyl glutaryl (HMG Co) reductase inhibitors or statin. The effect of simvastatin on TNF -alpha neutralizing antibody is that statin (3-hydroxy-3-methylglutaryl) coenzyme reductase inhibitors has the pleiotropic actions effect that can improve the survival of sepsis patients.

Objective: To prove the effect of simvastatin administration 0.03 mg, 0.06 mg and 0.12 mg PO on LPS intraperitoneal injected mice to the decrease of intraperitoneal macrophages’ phagocytosis capacity.

Methods: Experimental design research on post test only control group. The samples were 20 male mice type balb/c. Mice are divided into 4 groups, consisted of control group (without simvastatin injection), treatment group 1,2,3 consecutively administered simvastatin 0.03 mg; 0.06 mg; and 0.12 mg PO respectively. Initially these groups were injected intraperitoneal lipopolysaccharida 20 mg/kg.

Results: The mean capacity of macrophages’ phagocytosis for each groups: Control = 44,40+3,97; K 1 = 37,80+2,86; K 2 = 31,20+1,30; K 3 = 2,.00+4,30. The results of statistical tests between groups were shown significant differences between K1 with K3 and K4, between K2 with K3 and K4 (p<0,0,05). There were no significant differences between K1 dan K2, and between K3 and K4 (p>0.005).

Conclusion: The administration of simvastatin 0.03 mg, 0.06 mg and 0,12 mg PO show significant differences on the intraperitoneal macrophages’ phagocytosis capacity compared to the control group of mice with lipopolisakharida injection.

ABSTRAK

Latar Belakang : Simvastatin merupakan grup obat yang disebut sebagai hydroxy metyl glutaryl (HMG Co) reductase inhibitors). Efek simvastatin terhadap TNF -alpha neutralizing antibody bahwa Statins (3-hydroxy-3-methylglutaryl) coenzyme reductase inhibitors memiliki efek pleiotropic actions, yang mampu memperbaiki survival penderita sepsis.

Tujuan : Membuktikan efek pemberian simvastatin 0,03 mg, 0,06 mg dan 0,12 mg peroral pada mencit yang diberi LPS intraperitoneal terhadap penurunan kapasitas fagositosis makrofag intraperitoneal.

Metode : Penelitian eksperimental desain the post test only controlgroup. Sampel penelitian 20 ekor mencit balb/c jantan. Mencit dibagi dalam 4 kelompok, yaitu kelompok Kontrol (tidak diberi simvastatin), kelompok Perlakuan 1,2,3 berturut-turut diberi simvastatin 0,03 mg; 0,06 mg; dan 0,12 mg peroral.Sebelumnya masing -masing kelompok disuntikkan lipopolisakarida 10 mg/kgBB intraperitoneal.

Hasil : Rerata kapasitas fagositosis makrofag untuk masing-masing kelompok : Kontrol = 44,40+3.97; Perlakuan 1 = 37,80+2,86; Perlakuan 2 = 31,20+1,30; Perlakuan 3 = 23,00+4,30. Hasil uji statistik antar kelompok didapatkan perbedaan yang bermakna antara kelompok K1 dengan K3 dan K4, antara K2 dengan K3 dan K4 (p<0,0,05). Tidak terdapat perbedaan bermakna antara K1 dan K2, serta K3 dan K4. (p>0,0,05).

Kesimpulan : Pemberian simvastatin dosis 0,06 mg dan 0,12 mg peroral menunjukkan perbedaan bermakna pada penurunan kapasitas fagositosis makrofag intraperitoneal dibanding kontrol pada mencit yang diberi lipopolisakarida.
